PyroSequencing Procedure
The CORE offers two types of PYROSEQUENCING services
Pyrosequencing Full Service
Customer provides a clearly labeled document with the name of the sequence, and the sequence of the region of interest identifying the target region or SNP
The Core will design, order and optimize the primers, do the PCR and run an argarose gel to verify and do the pyrosequencing run.
Cost: One time project set up fee=$100 Primer design $25/primer set Pcr primers approx. $125/- (one plain and one biotinylated HPLC purified) Sequencing primer $25 Primer optimization: $150 for up to 4 pairs PCR charge + Labor $100/- per plate Pyro run $200/plate
Pyrosequencing Ready-to-Run Service
The customer is responsible for assay design, primer optimization, and PCR. The customer needs to provide the core the PCR product and an argarose gel image of the PCR with 1ul loaded on the gel and the sequencing primer.
When ordering primers please keep in mind that the primers need to be HPLC purified and the appropriate forward or reverse primer needs to be biotin labeled. Please also keep in mind that in order to get good results from your pyrosequencing run you need a very clean clearly visible and strong band on a gel.
Customer provides a clearly labeled document with the name of the sequence, and the sequence of the region of interest identifying the target region or SNP. Please also provide the pcr and sequencing primers clearly marked and the dispensation order. Customer provides minimum 50ul pcr product (do not purify the product) and the sequencing primer at 5micomolar conc. (minimum volume 500microliters per 96 well plate).
Cost: One time project set up fee= $100 Pyro run = $200/plate
