Pyrosequencing: A Rapid and Cost Effective Technology for Quantitative Genotyping, DNA Methylation, and Short-Read Sequencing Analysis
Pyrosequencing is the gold standard for accurate and highly sensitive mutation and CpG methylation analysis, combining quantitative analysis with highly reliable sequencing data. The patented “Sequencing-By-Synthesis” method uses no fluorescent labels, electrophoretic separations, or expensive dyes and provides the ability to easily and inexpensively analyze up to 96 samples simultaneously. By employing Pyrosequencing, researchers can precisely quantify extremely low levels of mutations or CpG methylation at a single or multiple sites in a single assay. The technology is particularly well suited for mutation and methylation detection within mixed sample populations such as tumor cells present with normal cells. Low frequency level mutations or methylation can therefore be determined in numerous genes linked to cancers such as; K-RAS, B-RAF, MLH1 and MGMT. This talk will highlight how Pyrosequencing works, application of the technology for rapidly analyzing SNPs, allele quantification, and DNA methylation, and many new advances in Pyrosequencing technology such as the PyroMark Q24 instrument, Assay Design Software 2.0, and a new family of Cancer RUO Mutation & DNA Methylation Analysis Tests which provide off the shelf simplicity for new assay validation.
